PENENTUAN KADAR TOTAL FENOLIK DAN FLAVONOID EKSTRAK ETANOL DAN FRAKSI DAUN TERAP (Artocarpus odoratissimus Blanco)

Daun terap (Artocarpus odoratissimus Blanco) merupakan salah satu tumbuhan tradisional yang memiliki beberapa aktivitas farmakologi karena mengandung metabolit sekunder berupa fenolik dan flavonoid. Penelitian ini bertujuan untuk mengetahui kadar total fenolik dan flavonoid pada dalam ekstrak dan fraksi daun terap dengan metode kolorometri. Metode penelitian penentuan kadar total fenolik menggunakan metode Folin Ciocalteu menggunakan pembanding asam galat dan penentuan kadar flavonoid menggunakan metode pembentukan kompleks AlCl3 dengan pembanding kuersetin dengan pengukuran secara spektrofotometri VU-Visible. Hasil Penelitian diperoleh hasil kadar total fenolik pada ekstrak etanol sebesar 154 mgGAE/g, fraksi n-heksana sebesar 124 mgGAE/g, fraksi etil asetat sebesar 106 mgGAE/g dan fraksi air sebesar 40 mg GAE/g. Hasil kadar total flavonoid pada ekstrak etanol sebesar 25 mg QE/g, fraksi n-heksana sebesar 32 mgQE/g, fraksi etil asetat sebesar 55 mgQE/g dan fraksi air sebesar 28 mgQE/g. Berdasarkan hasil tersebut dapat disimpulkan bahwa kadar total fenolik lebih besar dibandingkan kadar flavonoid di ekstrak dan fraksi.
Terap leaves (Artocarpus odoratissimus Blanco) is one of traditional plant have several pharmacological activities because of secondary metabolite as know phenol and flavonoids. This study aimed to determine the total content of phenol and flavonoids in extract and applied leaf fraction determined by colorimetric method. Research method for determining total phenol content using the Folin Ciocalteu method using gallic acid as standard and determining total flavonoids content using complex formation method with AlCl3 using quersetin as standard by measurement with UV-Vis spectrophotometry. The results of the total content phenol and flavonoids showed that the ethanol extract of 154 mgGAE/g, n-hexane fraction of 124 mgGAE/g, ethyl acetate fraction of 106 mgGAE/g, water fraction of 40 mg GAE/g. Determination of total flavonoids using the AlCl3 complex formation method, obtained results in the ethanol extract of 25 mg QE/g, n-hexane fraction of 32 mgQE/g, ethyl acetate fraction of 55 mgQE/g, water fraction of 28 mgQE/g. Based on results, it can be concluded that the total phenol content is higher than total flavonoids content.
