ISOLASI DAN UJI AKTIVITAS ANTIOKSIDAN SENYAWA METABOLIT SEKUNDER EKSTRAK DCM DAUN UNGU (Graptophyllum pictum L. (Griff)

Telah dilakukan isolasi senyawa metabolit sekunder dari ekstrak DCM daun ungu (Graptophillum pictum Griff). Penelitian ini bertujuan untuk mengisolasi senyawa metabolit sekunder dari ekstrak DCM daun ungu dan mengevaluasi aktivitas antioksidannya. Ekstraksi dilakukan menggunakan metode maserasi bertingkat, dilanjutkan dengan pemisahan melalui kromatografi kolom flash dan pemurnian dilakukan dengan cara dekantasi dan rekristalisasi sehingga menghasilkan isolat dari fraksi gabungan yang diberi label isolat GPD-1. Isolat dikarakterisasi secara organoleptis yang diperoleh berupa padatan serbuk berwarna putih, berbau tajam, titik leleh 210-212℃, larut dalam DMSO dan metanol yang ditambahkan etil asetat. Identifikasi isolat GPD-1 menggunakan pereaksi penampak noda anisaldehid menunjukkan isolat GPD-1 merupakan golongan steroid dan hasil karakterisasi FT-IR, 1H-NMR, 13C-NMR dan HSQC menunjukkan golongan steroid yang diprediksi dengan nama Stigmasta-5,22-dien-3β-O-D-glucopyranoside. Pengujian Aktivitas antioksidan dilakukan dengan metode DPPH untuk menentukan nilai IC50 dari ekstrak dan isolat. Hasil pengujian aktivitas antioksidan menunjukkan bahwa ekstrak DCM daun ungu dan isolat GPD-1 tidak memiliki potensi sebagai aktivitas antioksidan dengan hasil nilai IC50 yang diperoleh yaitu > 1000 µg/mL.
Secondary metabolite compound has been isolated from purple (Graptophyllum pictum Griff) leaf DCM extract. This research aims to isolate secondary metabolite compound from purple leaf dichloromethane extract and evaluate its antioxidant activity. Extraction is carried out using the multi-layered maceration method, followed by separation through flash column chromatography and purification is carried out by decantation and recrystallization so as to produce isolates from the combined fraction which are labeled GPD-1 isolate. Isolates are characterized organoleptically which are obtained in the form of a white powdery solid, sharp odor, melting point of 210-212℃, soluble in DMSO and methanol with added ethyl acetate. identification of GPD-1 isolate using anisaldehyde staining reagent shows that GPD-1 isolate is a steroid group and the results of FT-IR, 1H-NMR, 13C-NMR and HSQC characterize the steroid group predicted under the name Stigmasta-5,22-dien-3β-O-D-glucopyranoside. Antioxidant Activity Testing is carried out with the DPPH method to determine the IC50 value of extracts and isolates. The results of antioxidant activity testing show that DCM extract of purple leaves and GPD-1 isolate do not have the potential as antioxidant activity with the result of IC50 values obtained > 1000 µg/mL.
